ABSTRACT

Ethyl-3 Stephania hernandifolia Walp. alkaloid is chemical substance isolated from tuber of S. hernandifolia Walp. plant. It was presumed as substance consisting many medicinal importances but informations about its negative effects were still restricted. This research was conducted to analyse about its acute toxicity effects on hematological values of albino mice (Mus musculus L.). Male albino mice were exposed to alkaloid substance at several dose levels such as 0, 25, 50, and 75 mg/kg of body weight (bw) for 24, 48, and 72 hours respectively. The blood was collected at the end of each exposure period and analysed hematologically for red blood cell count (RBC), white blood cell count (WBC), hematocrite or packed cell volume (PCV), hemoglobin concentration (Hb), plasm protein concentration (PPC), and blood indices. Ethyl-3 Stephania hernandifolia Walp. alkaloid caused acute toxicity effects on hematological values of mice expressed in decline of RBC, PCV, Hb, and PPC; and increasing of Mean Corpuscular Volume (MCV), Mean Corpuscular Hemoglobin (MCH), and WBC. Significant toxicity effects commonly occurred at medium dose level (50 mg/kg bw) and high dose level (75 mg/kg bw).

Key words: acute toxicity, alkaloid, hematological value
